#C198 Synergistic Effect of Tahitian Noni Juice (TNJ) and Methylsulfonylmethane (MSM) on Mammary Breast Cancer Prevention at the Initiation Stage of Chemical Carcinogenesis Induced by DMBA in Female Sprague-Dawley (SD) Rats.

Univ. of Illinois College of Medicine, Rockford, IL. Our previous studies have demonstrated that the natural nutritional supplements TNJ and MSM have a cancer preventive effect at the initiation stage of chemical carcinogenesis in a mammary breast cancer model.

In this study, we examined whether TNJ and MSM have a synergistic effect on mammary breast cancer prevention at the initiation stage of chemical carcinogenesis induced by DMBA in female SD rats.

The experiment was started at the 35th postnatal day with water in an age-matched control group, a DMBA group, a 5% TNJ group, a 5% MSM group, and a combination of 5% TNJ plus 5% MSM group. DMBA (25 mg/kg) was administrated by mouth at the 50th postnatal day in the DMBA, TNJ, MSM, and combination groups.

TNJ and MSM were continuously supplied for an additional 90 days after DMBA administration. All the animals were sacrificed at the 8th month after DMBA administration to examine the pathological changes in the mammary glands by light microscopy.

Compared to the controls, the DMBA treated group showed a variety of lesions, including epithelial hyperplasias (12.5%), benign tumors (25%), and in-situ carcinomas (25%).

No benign tumors or carcinomas were observed in the TNJ, MSM, and the combination groups, which showed only 60%, 75%, and 37.5% mild hyperplasia, respectively.

Our results indicate that TNJ and MSM may prevent mammary breast cancer at the initiation stage of chemical carcinogenesis.

A synergistic cancer preventive effect was observed in the combination group. The mild hyperplasia in the combination group was decreased 50% and 37.5%, respectively, compared with that of the TNJ and MSM groups.

TNJ plus MSM may be a new regime to prevent mammary breast cancer in humans.

The mechanisms for this new discovery needs further study.
